ABSTRACT
Cabbage (Brassica oleracea var. capitata L.) is one of agricultural important crop that has some cultivars. Productivity of the crop is affected by some factors. One of disadvantage that can decrease the productivity is presence of disease caused by fungal pathogen infection. Some cabbage cultivars had been identified to possess defence system for pathogen infection. β-1,3-endoglukanase is one of enzyme which has a function to defence against pathogen fungal infection. This enzyme catalyzes the hydrolysis of β-1,3-glukan which is a major component of cell wall of most fungi and it has a molecule weight of 33 kDa. The hydrolyze yields elicitors and it then induces production of phytoalexins antifungal. This research was conducted for early identification of β-1,3-endoglukanase protein based on its molecular weight in SDS-PAGE (Sodium Dodecyl Sulphonat Polyacrylamide Gel Electrophoresis). The identification was carried out after cabbage cultivars (KK Cross, Gloria Osena, Ishito 3, Shinjuku, Rotan Osena, dan Investor 369) were infected by four fungal pathogens that are Penicillium sp., Mucor sp., Aspergillus sp., and Alternaria sp. One week after infection, the presence of necrotic on surface of leaves was observed. Result of necrotic observation showed that some of the cabbage cultivar was susceptible to the fungal pathogen infections, that were KK Cross and Shinjuku cultivars. However, Gloria Osena, Ishito 3, Rotan Osena, and Investor 369, were more resistance to fungal pathogen infection than KK Cross and Shinjuku. The SDS-PAGE analysis showed that the cultivars which were infected by Alternaria sp. and Aspergillus sp. could not produce β-1,3-endoglukanase protein band. These results showed that cabbage cultivars were susceptible when they were infected by Alternaria sp. and Aspergillus sp.
